چکیده

Neutralizing monoclonal antibodies were developed against strains of infectious hematopoietic necrosis virus (IHNV) from steelhead trout Salmo gajrdneri in the Deschutes River of Oregon, chinook salmon Oncorhynchus tshawytscha in the Sacramento h v e r of California, and rainbow trout Salmo gairdneri reared in the Hagerman Valley of Idaho. USA. These antibodies were tested for neutralization of 12 IHNV isolates obtained from salmonids in Japan, Alaska, Washington, Oregon, California, and Idaho. The antibodies recognized antigenic variants among the isolates and could be used to separate the viruses into 4 groups. The members of each group tended to be related by geographic area rather than by source host species, virulence, or date of isolation.

Rapid detection of antigenic diversity of Akabane virus isolates by dot immunobinding assay using neutralizing monoclonal antibodies.

Akabane (AKA) virus is an arthropod-borne virus belonging to the Simbu group of the genus Bunyavirus. Neutralizing monoclonal antibodies (MAbs) against AKA virus were prepared, and the neutralizing epitopes of the virus were defined by competitive binding assay. Five distinct antigenic domains were identified and were designated A, B, C, D, and E. Domains A and C consisted of two epitopes each....

Mapping the neutralizing epitopes on the glycoprotein of infectious haematopoietic necrosis virus, a fish rhabdovirus.

Twelve neutralizing monoclonal antibodies (MAbs) against the fish rhabdovirus, infectious haematopoietic necrosis virus (IHNV), were used to select 20 MAb escape mutants. The nucleotide sequence of the entire glycoprotein (G) gene was determined for six mutants representing differing cross-neutralization patterns and each had a single nucleotide change leading to a single amino acid substitutio...

Detection of spring viraemia of carp virus (SVCV) with monoclonal antibodies

Monoclonal antibodies (MAbs) against spring viraemia of carp virus (SVCV), a severe disease in cyprinid fish, were prepared. Nine MAbs were characterised using Western blotting (WB) where all reacted with glycoprotein G, except for MAb 2E1, which showed no reactivity in WB. All nine MAbs showed specificity in an immunoperoxidase test. In ELISA assays, their titres ranged between 1:32 000 and 1:...

Antigenic variants of rabies virus

The authors studied nineteen street virus isolates from different regions of Canada using either anti-nucleocapsid and anti-glycoprotein monoclonal antibodies or cross-protection tests. This study only allowed us to recognize three groups of viruses with different nucleocapsid patterns, and no difference, as far as protection with a Pitmann-Moore vaccine is concerned, between four of these stra...

Host cell selection of antigenic variants of foot-and-mouth disease virus.

Foot-and-mouth disease virus (FMDV) A22 Iraq 24/64 adapted to grow in BHK monolayer cells induced antibodies which neutralized many isolates belonging to the A serotype. Plaque-purified virus isolated from this stock also induced broadly reactive antibodies, showing that this property is not due to the combined response to a mixture of variants in the original stock virus. However, viruses obta...
